چکیده
Abstract Background In inflammatory bowel disease (IBD), including Crohn’s (CD) and ulcerative colitis (UC), CD4+ T cell-responses to unknown microbial antigens drive intestinal inflammation. Both hypo- or hyperactive anti-microbial innate immunity may underlie these pathogenic T-cell responses. Previously, IgG responses Lachnospiraceae flagellins were detected in spontaneously colitic mice with an immune defect adult CD patients complicated disease. However, whether occur therapy-naïve pediatric they relate hyperresponsiveness is unknown. We hypothesize that, at diagnosis, a subgroup of has elevated anti-Lachnospiraceae flagellin concomitant flagellin-specific associating insufficient immunity. Methods Plasma reactivity 20 from that colonize mouse human gut was measured IBD (CD n=49; UC n=16), non-IBD (n=12) age-matched healthy controls (HC n=17). divided two groups: ‘flagellin-hi’ (elevated 11-20 flagellins) ‘flagellin-lo’ 0-10 flagellins), whose immunological clinical profiles compared. Results The proportion flagellin-hi higher (41%) compared (6%), (8%) HC (6%). patients, activity score, CRP ESR did not reactivity. Compared flagellin-lo had increased frequencies gut-homing (CD38+CD62Lneg) circulating cells. Crucially, high Although anti-flagellin associated flagellin-specific- gut-homing- the circulation, it very limited cell infiltration, epithelial damage phosphorylated NFκB colonic biopsies on histology. Moreover, plasma concentration EN-RAGE, neutrophil-derived protein, decreased -lo patients. Conclusion sum, Lachnospiraceae-derived flagellins. While have cells, lower histological activation, possibly suggesting activation and/or infiltration. Together, our data raise question enhanced adaptive response be driven by local

Elevated Adaptive Immune Responses Are Associated with Latent Infections of Wuchereria bancrofti
In order to guarantee the fulfillment of their complex lifecycle, adult filarial nematodes release millions of microfilariae (MF), which are taken up by mosquito vectors. The current strategy to eliminate lymphatic filariasis as a public health problem focuses upon interrupting this transmission through annual mass drug administration (MDA). It remains unclear however, how many rounds of MDA ar...
متن کاملElevated EBNA1 Immune Responses Predict Conversion to Multiple Sclerosis
Objective—The aims of the study were to determine the immune responses to candidate viral triggers of multiple sclerosis (MS) in patients with clinically isolated syndromes (CIS), and to evaluate their potential value in predicting conversion to MS. Methods—Immune responses to Epstein-Barr virus (EBV), human herpesvirus 6, cytomegalovirus (HCMV), and measles were determined in a cohort of 147 C...

Objective(s): Bacterial translocation in patients with cirrhosis is an important triggering factor for infections and mortality. In the bile duct ligation (BDL) model, crucial players of bacterial translocation are still unknown. This study aims to determine the interrelation between microbiome composition in the colon, mesenteric lymph nodes, and liver, as well as the...
متن کاملDNA methylation-associated colonic mucosal immune and defense responses in treatment-naïve pediatric ulcerative colitis
Inflammatory bowel diseases (IBD) are emerging globally, indicating that environmental factors may be important in their pathogenesis. Colonic mucosal epigenetic changes, such as DNA methylation, can occur in response to the environment and have been implicated in IBD pathology. However, mucosal DNA methylation has not been examined in treatment-naïve patients. We studied DNA methylation in unt...
متن کاملThe Modulation of Adaptive Immune Responses by Bacterial Zwitterionic Polysaccharides
The detection of pathogen-derived molecules as foreign particles by adaptive immune cells triggers T and B lymphocytes to mount protective cellular and humoral responses, respectively. Recent immunological advances elucidated that proteins and some lipids are the principle biological molecules that induce protective T cell responses during microbial infections. Polysaccharides are important com...
